Acest Sfint Sinod Ecumenic a fost convocat la Constantinopol în anul 553, sub domnia împaratului lustinian cel Mare. La acest Sinod au fost condamnate toate feluritele erezii ale monofizismului, precum si scrierile eretice ale lui Teodor de Mopsuestia si Teodoret din Cir, si învatatura gresita a lui Origen (împotriva învierii mortilor).
