El a fost Episcop al cetăţii Taiski din Thabaida Egiptului. El a avut de suferit muceniceşte întru apărarea Ortodoxiei Credinţei. Ereticii 1-au mutilat, scoţîndu-i un ochi şi tăindu-i piciorul stîng. El a fost unul dintre Sfinţii Părinţi ai Sinodului I Ecumenic [de la Niceea, din anul 325], şi dintre cei mai combativi împotriva ereziei lui Arie. Sffntul împărat Constantin cel Mare a avut pentru el un respect adînc, şi îl săruta adesea pe ochiul care îi lipsea, cel care fusese scos întru apărarea Ortodoxiei. Acest Sfînt Pafnutie a stat cu putere împotriva reprezentanţilor apuseni la Sinod, care voiau ca preoţii de mir să fie şi ei bărbaţi necăsătoriţi. Sfîntul Pafnutie Mărturisitorul a fost feciorelnic toată viaţa lui.
