Ὁ Ἅγιος Ἀναστάσιος ἀσκήτεψε στὸ ὄρος Σινᾶ περὶ τὶς ἀρχὲς τοῦ 6ου αἰῶνος μ.Χ., γι’ αὐτὸ καὶ ἀποκαλεῖται Σιναΐτης. Ἀπὸ τὸ Σινᾶ μετέβη στὴν Ἀντιόχεια, ὅπου ἔγινε ἀποκρισάριος τῆς Ἐκκλησίας τῆς Ἀλεξανδρείας. Ὅταν πέθανε ὁ Ἐπίσκοπος Ἀντιοχείας Δόμνος Γ’ (546 – 561 μ.Χ.), κλήθηκε ἀπὸ τὸν κλῆρο καὶ τὸν λαὸ σὲ διαδοχὴ αὐτοῦ. Ὁ αὐτοκράτορας Ἰουστίνος (565 – 576 μ.Χ.), μὲ τὴν πρόφαση ὅτι ὁ Ἅγιος κατασπατάλησε τὴν ἐκκλησιαστικὴ περιουσία, τὸ ἐξόρισε τὸ ἔτος 571 μ.Χ. στὰ Ἱεροσόλυμα. Ἐκεῖ ὁ Ἅγιος παρέμεινε μελετώντας καὶ συγγράφοντας μέχρι τὸ ἔτος 593 μ.Χ., ὁπότε καὶ ἐπανῆλθε στὸ θρόνο του καὶ κοιμήθηκε κατὰ τὸ ἔτος 599 μ.Χ. Περὶ τοῦ τέλους αὐτοῦ, τὸ ὁποῖο οἱ Συναξαριστὲς ἀναφέρουν ὡς μαρτυρικό, δὲν ἔχουμε θετικὲς πληροφορίες.
