Ὁ Ὅσιος Ἀντώνιος γεννήθηκε πρὸ τοῦ 830 μ.Χ. στὴν Κωνσταντινούπολη. Σὲ μικρὴ ἡλικία ἔγινε μοναχὸς καὶ ἀναδείχθηκε «ἀνὴρ ὅσιος, ἐστεμμένος μὲ ἀρετὲς καὶ ναὸς οἴκτου». Χειροτονήθηκε Πρεσβύτερος καὶ μὴ θέλοντας νὰ ἐγκαταλείψει τὸν μοναχικὸ βίο ἵδρυσε τὴ μονὴ τοῦ Καλλίου ἢ Καλλέως. Ὁ Ὅσιος ἐργάσθηκε πολὺ γιὰ τὴν ἑνότητα τῆς Ἐκκλησίας καὶ τὴν κατασίγαση τῶν παθῶν μεταξὺ τῶν ὀπαδῶν τῶν Πατριαρχῶν Ἰγνατίου καὶ Φωτίου καὶ ἔγινε περιλάλητος γιὰ τὴν φιλανθρωπία καὶ τὴν ἐλεημοσύνη του. Τὸ ἔτος 893 μ.Χ. ἐξελέγη Πατριάρχης Κωνσταντινουπόλεως καὶ ἐξακολούθησε νὰ αὐξάνει στὴν πνευματικὴ προκοπή, τὴν εὐσέβεια καὶ τὴ φιλαδελφία.
