Ο Άγιος Μάρκος, ήταν Κρητικός και έμενε στη Σμύρνη, όπου από μικρό παιδί εξισλαμίστηκε βίαια από τούς Τούρκους. Αργότερα πήγε στην Κωνσταντινούπολη και μαθήτευσε κοντά στο σοφό και πολυμαθή διδάσκαλο, Μελέτιο Συρίγο. Επανήλθε στη Σμύρνη, όπου κήρυξε δημόσια το Χριστιανισμό. Για το λόγο αυτό συνελήφθη και αφού με σθένος αρνήθηκε να προδώσει την πίστη του, βασανίστηκε σκληρά και τέλος αποκεφαλίστηκε στις 14 Μαΐου 1643 μ.Χ. στη Σμύρνη. Το Ιερό του λείψανο παρέλαβαν οι χριστιανοί και το έθαψαν με τιμές στον ναό της Αγίας Φωτεινής Σμύρνης. Το μαρτύριο του Νεομάρτυρα αυτού διηγήθηκε ο Ισουΐτης Ισαάκ Α'.
