Η καταγωγή του Οσίου Ρωμανού ήταν από την Κιλικία, και συγκεκριμένα από την πόλη Ρώσο (αρχαία πόλη της Συρίας στον Ισσικό κόλπο - Αλεξανδρέτας - που βρίσκεται κοντά στις Κιλικίες Πύλες). Ο πνευματικός του όμως αγώνας, έγινε στα μέρη της Αντιόχειας. Εκεί, στους πρόποδες ενός βουνού έκτισε ένα μικρό κελί, που το χρησίμευε σαν μελετητήριο και συγχρόνως σαν ασκητήριο. Ήταν τύπος εγκρατής και στους τρόπους του απλός, γεμάτος ταπεινοφροσύνη και πραότητα.
