Als Sohn der Adligen Konstantin und Feodosija, der späteren Nonne Fekla, aus Mzensk um 1460 in Perejaslavl' -Zalesskij geboren, wurde er auf den Namen Dimitrij getauft. Er empfing in jungen Jahren die Mönchsweihe auf den Namen Daniil im Kloster des hl. Pafnutij von Borovo und führte sein geistliches Leben unter der Leitung des hl. Levkij von Volokolamsk (Gedenktag: 17. (4.) August). Später kam er in das Nikita-Kloster zu Perejaslavl'. Seine besondere Tugend bestand darin, dass er mittellose Tote begrub. Bei dem Friedhof gründete er selbst 1508 ein Kloster zu Ehren der Heiligsten Dreiheit. Besonders ehrte ihn Zar Vasilij III., der ihn zum Erzieher seiner Söhne Ioann und Georgij machte. Daniil starb am 7. April 1540.
