Paulinus stammte aus einer römischen Senatorenfamilie, er war der Vetter der älteren Melania von Rom. Paulinus erhielt eine umfassende wissenschaftliche Bildung und wurde 381 Statthalter in der Region Kampanien. Nach dem frühen Tod seines Sohnes lebte er mit seiner Frau zurückgezogen in den Pyrennäen. 394 wurde er in Barcelona zum Priester geweiht, dann gründete zusammen mit seiner Frau eine streng asketische klösterliche Gemeinschaft in Nola. 409 (oder 411) wurde er unter dem Jubel des Volkes zum Bischof gewählt. Er sorgte sich um die Armen und linderte die durch den Einfall der Westgoten unter Alarich entstandene Not.
