Amphilochius, ein Vetter von Gregor von Nazianz und Freund von Basilius dem Großen, studierte Jura und Rhetorik. Er war Rechtsanwalt in Konstantinopel - dem heutigen Ístanbul - und gab seinen Beruf zugunsten eines asketischen Lebens auf. Basilius veranlasste um 375 seine Wahl zum Bischof von Ikonium - dem heutigen Konya - und zum Metropoliten der Provinz Lykaonien veranlasste. Er bekämpfte den in der Provinz Asia stark verbreiteten Arianismus und den Manichäismus. Gegen die Irrlehrer setzte er auch weltliche Macht ein und überzeugte den zunächst widerwilligen Kaiser Theodosius, den Arianismus zu verbieten.
