起初，阿纳托利乌斯在亚力山大一个教堂中任司祭职，在牧首弗拉维安去世后，阿纳托利乌斯于公元449年被选为君士坦丁堡牧首。在他管理教会期间，公元451年在卡尔西顿举行的普世大公会议上，君士坦丁堡和罗马两个牧首被确立拥有同等的地位。阿纳托利乌斯为维护东正教的纯正进行了艰苦的斗争，多次遭受异端的迫害，最后于公元458年教宗大莱翁统治期间被异端杀害。阿纳托利乌斯管理教会九年的时间，最后得以进入上帝的国度，与众圣人同享荣耀。
