Acest sfint sinod ecumenic a fost convocat în timpul domniei împaratului Teodosie cel Mare la Constantinopole, la anul 381. Scopul lui principal a fost anatematizarea lui Macedon, ereticul luptator împotriva Duhului Sfînt. Macedon, Patriarh al Constantinopolelui, învata mincinos ca Duhul Sfînt este o creatie a lui Dumnezeu iar nu Persoana Dumnezeiasca (Ipostas Dumnezeiesc), si prin urmare nu este de o fiinta si de o închinare cu Tatal si cu Fiul, Unul din Sfînta Treime. Macedon a fost dat anatemei la acest sfînt sinod, si astfel învatatura despre Duhul Sfînt a fost adaugata Simbolului de credinta niceean.
